چکیده

The transcription factor hypoxia-inducible factor (HIF)-1 is an important mediator of hypoxic adaptation of tumor cells and controls several genes that have been implicated in tumor growth. Oxygen-dependent degradation of HIF-1a, the regulatory subunit, requires binding to the von Hippel Lindau (VHL) protein. Because functional inactivation of the VHL tumor suppressor gene occurs in up to 70% of clear cell renal carcinomas, we investigated whether this results in overexpression of HIF-1a and its target genes. Immunoblotting revealed increased expression of HIF-1a in 24 of 32 (75%) clear cell renal carcinomas but only 3 of 8 non-clear cell renal tumors. Somatic mutations of the VHL gene were detected only in clear cell renal carcinomas that overexpressed HIF-1a. None of the HIF-1a-negative tumors displayed a VHL mutation. The level of HIF-1a mRNA was not different between tumors and adjacent kidney tissue. Immunohistochemistry revealed distinct patterns of nuclear staining for HIF-1a, depending on histological type and overall abundance of HIF-1a. In those clear cell renal carcinomas that showed increased expression on immunoblots, HIF-1a was expressed in almost all cells. In the remaining clear cell and in non-clear cell tumors, staining was focal; these different patterns thus were compatible with genetic stabilization in contrast to microenvironmental stimulation of HIF-1a as the primary mechanism. The mRNA expression of two known target genes of HIF-1a, vascular endothelial growth factor and glucose transporter 1, increased progressively with increasing amounts of HIF-1a in tumor extracts. In addition, glucose transporter 1 protein levels correlated with HIF-1a abundance. In conclusion, the data provide in vivo evidence for a constitutive up-regulation of HIF-1a in the majority of clear cell renal carcinomas, which leads to more widespread accumulation of this transcription factor than hypoxic stimulation. These observations are most likely linked to functional inactivation of the VHL gene product. Increased expression of HIF-1a is associated with alterations in gene expression patterns that are likely to contribute to tumor phenotype and progression.

Angiogenesis, as the process of new vessel formation from pre-existing vessels is dependent on a delicate equilibrium between endogenous angiogenic and antiangiogenic factors. However, under pathological conditions, this tight regulation becomes lost which can result in the formation of the different diseases such as cancer. Angiogenesis is a complex process that includes many gene products tha...

Blockade of Hypoxia: The Impact on Tumor Growth in an Experimental Tumor Model

Background: Tumor microenvironment is an active factor participating in immunoregulation, thereby preventing immunosurveillance and limiting the efficacy of anticancer therapies. Hypoxia as a major characteristic of solid tumors causes the expression of Hypoxia-Inducible Factor-1α (HIF-1α). This is a transcription factor that mediates hypoxic responses of tumor cells and involves in the express...

Investigation of the effect of sublethal concentrations of endosulfan on gene expression of hypoxia-inducible factors (HIF-1 α and HIF-2 α) and vascular endothelial growth factor (VEGF) in the Persian sturgeon fingerlings (Acipenser persicus)

In this study, the effects of sublethal concentrations of the Endosulfan on gene expression of hypoxia-inducible factor (HIF-1 α, HIF-2 α) and vascular endothelial growth factor (VEGF) in the gill tissue of Persian sturgeon fingerling (Acipenser persicus) were studied. A total of 360 sturgeons (average weight of 2 ± 0.12 g) exposed to concentrations of 10, 20 and 40 micrograms per liter and VEG...

Human activated macrophages and hypoxia: a comprehensive review of the literature

Macrophages accumulate in poorly vascularised and hypoxic sites including solid tumours, wounds and sites of infection and inflammation where they can be exposed to low levels of oxygen for long periods. Up to date, different studies have shown that a number of transcription factors are activated by hypoxia which in turn activate a broad array of mitogenic, pro-invasive, pro-angiogenic, and pro...

Identification of small molecule inhibitors of hypoxia-inducible factor 1 transcriptional activation pathway.

Hypoxia-inducible factor 1 (HIF-1) is a master regulator of the transcriptional response to oxygen deprivation. HIF-1 has been implicated in the regulation of genes involved in angiogenesis [e.g., vascular endothelial growth factor (VEGF) and inducible nitric oxide synthase] and anaerobic metabolism (e.g., glycolytic enzymes). HIF-1 is essential for angiogenesis and is associated with tumor pro...
